A poster presented at the 2025 Society of Dermatology Physician Associates Summer Dermatology Conference highlighted the effectiveness of dupilumab (Dupixent; Regeneron Pharmaceuticals and Sanofi) in providing sustained, long-term flare control in adult patients with moderate to severe atopic dermatitis (AD).

The analysis assessed data from the phase 3 SOLO 1/2 (NCT02277743; NCT02277769)

 and SOLO-CONTINUE extension (NCT02395133)

 trials. The investigation focused on the extent to which dupilumab can prevent flare episodes over a full year of continuous treatment. 

Unlike previous studies combining dupilumab with topical corticosteroids, the present analysis centered solely on dupilumab administered alone, without adjunct therapies, to determine its standalone efficacy and safety in maintaining disease control.

Participants in the study had already responded favorably to a 16-week initial treatment phase, receiving treatment with dupilumab 300 mg once per week or once every 2 weeks. Those who demonstrated significant improvement, defined by achieving an Investigator Global Assessment score of 0/1 (indicative of complete or almost complete clearance) or at least a 75% improvement in Eczema Area and Severity Index, were eligible for extension into the next phase of the trial.

During this phase, more than 160 adults were randomly assigned into different maintenance dose groups: 300 mg every 2 weeks, 300 mg every 4 weeks, and 300 mg every 8 weeks. The study then tracked the occurrence of flares, defined as worsening requiring treatment escalation, and analyzed the duration until the first flare event.

Approximately 80% of patients (equivalent to 8 of every 10 patients) across all dosing schedules remained well controlled, with no disease flares observed throughout the duration of the 1-year follow-up.

The duration of time from baseline to first observed flare was significantly delayed in patients treated with dupilumab, regardless of whether they received injections every 2, 4, or 8 weeks.

The safety profile was consistent with known data on dupilumab, with most adverse events being mild to moderate in nature. Serious adverse events were rare, and no treatment-related deaths occurred.

"Dupilumab monotherapy over 1 year prevented flares in 8 of 10 patients regardless of the maintenance dose regimen," wrote poster authors Simpson et al.

The findings suggest that dupilumab monotherapy is highly effective in maintaining disease control over an extended period, emphasizing its role as a sustainable long-term solution in AD management.

Dupilumab Maintains Long-Term Flare Control Across Dosing Regimens in Atopic Dermatitis

Alopecia areata affects approximately 2% of the global population, translating to around 160 million individuals worldwide. In the United States, about 6.7 million people have experienced or will experience this condition, with an estimated 700,000 currently affected. This autoimmune disorder impacts both children and adults, leading to varying degrees of hair loss. The data underscores the prevalence of alopecia areata and highlights the need for continued research and awareness to better understand and manage this condition.

In early 2025, significant changes in federal research funding policies have introduced uncertainty for the medical research community, including dermatology. These developments may impact clinical trials, translational research, and the training of future physician-scientists.

The National Institutes of Health (NIH) implemented a policy capping indirect cost reimbursements at 15%, down from previous rates that often exceeded 50%.

 Although a federal judge temporarily blocked this policy following legal challenges, many institutions have already initiated hiring freezes, reduced graduate admissions, and paused research projects.

Effective January 25, 2025, the NIH introduced a simplified peer review framework for research project grants, fellowships, and training grants.

 These changes aim to streamline the application process and promote fairness by focusing on key assessments such as candidate potential and research training plans.

The NIH has ended funding for programs related to climate change and health, reflecting a shift in research priorities.

 Additionally, Notices of Funding Opportunities related to diversity, equity, inclusion, and health disparities have been removed, raising concerns about the future direction of NIH-funded research.

Derm Dispatch: Melanoma Doesn't Wait -- Neither Should You

, Renata Block, PA-C, spoke with Brittanny Groover, a melanoma survivor and passionate advocate for skin cancer awareness. Groover, now in her late twenties, shared the deeply personal story of how she developed melanoma after years of frequent tanning, a habit formed in her teen years and encouraged by her environment. Growing up with her mother—who also loved tanning—Groover spent countless hours in the sun and later, in tanning beds. Even after beginning regular skin checks as a young adult, her belief that "it won’t happen to me" persisted until the moment it did.

Her diagnosis came after she noticed a mole on her shin growing in size and changing in appearance. Although she had an established history of annual dermatology visits, the COVID-19 pandemic disrupted her routine, delaying her next skin check. When she finally returned to the clinic, her dermatologist immediately identified the mole as melanoma—later confirmed by biopsy. She underwent a wide local excision with a skin graft, followed by a sentinel lymph node biopsy that revealed the cancer had begun to spread. Brittany then endured a year of immunotherapy (Keytruda), which led to intense adverse effects such as colitis and pancreatitis—complications she now says were worse than the cancer itself.

Now cancer-free, Groover has become a powerful voice for melanoma awareness, particularly among younger audiences. She shares her journey unfiltered on social media to combat the perception that skin cancer is minor or merely cosmetic. Her goal is to show that melanoma is not “just skin cancer” and can have life-altering consequences. She encourages clinicians to engage young patients through real-life stories and hands-on education, rather than brochures alone. Teaching teens how to perform skin checks, understand sun protection, and recognize warning signs could make all the difference.

Through her work with initiatives like “Don’t Fry Day” and the National Council on Skin Cancer Prevention, Groover is helping reshape how young people understand skin health. Her message is clear: melanoma doesn’t discriminate by age, and prevention must start early.

Derm Dispatch: Melanoma Doesn’t Wait—Neither Should You

At the 2025 Society of Dermatology Physician Associates (SDPA) Summer Dermatology Conference, a poster presented by Lebwohl et al offered a comprehensive update on the long-term safety of brodalumab (Siliq; Bausch Health Companies Inc).

Now in its seventh year of postmarketing surveillance in the US, brodalumab’s risk profile remains stable, with no new safety signals and low rates of adverse events (AEs), including those of special clinical concern such as suicidality, fungal infections, and major adverse cardiac events (MACEs). The pivotal trials for brodalumab did not exclude individuals with psychiatric comorbidities or suicidality risk factors, and no causal relationship has been established between the drug and suicidality.

The findings serve as a follow-up to the team’s previously published 6-year pharmacovigilance report

 and further support the safety of this IL-17A receptor antagonist, which was FDA approved in 2017 for moderate to severe plaque psoriasis in adults who have experienced treatment failure or lost response to other systemic therapies.

The pharmacovigilance data analyzed were submitted to Ortho Dermatologics by patients and health care providers from August 15, 2017, through August 14, 2024. The data set captured safety information from 5449 patients treated with brodalumab, accounting for 7845 patient-years of exposure.

During the 7-year surveillance period, 49.9% of patients (2719 of 5449) treated with brodalumab experienced at least 1 reported AE, with approximately a quarter of these cases submitted by health care providers. Arthralgia was the most frequently reported AE, occurring in 140 patients, consistent with the rate observed in year 6.

Other commonly reported AEs included headache in 58 patients, fatigue in 54, injection site reactions in 43, and myalgia in 40. Diarrhea and nausea were reported in 37 and 32 patients, respectively. No new cases of neutropenia were observed, and only 1 new tinea infection was reported during the most recent year.

With respect to psychiatric safety, no completed suicides were reported over the full 7-year period. Only 1 suicide attempt occurred, which was reported during year 3. Depression was documented in 60 patients, on par with data from the previous year’s analysis.

Infectious complications were also monitored. A total of 118 serious infections were reported. Only 4 of these events were considered related to brodalumab. Two new 

 infections were documented in year 7, and no new COVID-19–related deaths occurred. Overall, the rate of fungal infections remained low at 0.17 per 100 patients, with no new invasive fungal infections reported since year 6. Inflammatory bowel disease was infrequent, with only 3 cases reported over the entire 7-year period, and no additional cases recorded in year 7. 

Malignancy events were reported in 55 patients, totaling 64 cases. These included neoplasms of the right ovary, colorectal cancer, 2 cases of basal cell carcinoma, 1 case of squamous cell carcinoma, and isolated reports of renal and prostate cancers.

Regarding cardiovascular risk, there were no new adjudicated MACEs reported in year 7. Across the entire surveillance period, 13 adjudicated MACEs were reported. Of these 13 patients, 92% had preexisting cardiovascular risk factors. Furthermore, more than half of the patients (54%) continued brodalumab therapy following their cardiac event.

"Pharmacovigilance data throughout 7 years are consistent with the established safety profile of brodalumab observed in long-term clinical trials and previous US pharmacovigilance reports, with no completed suicides and a low fungal infection rate," Lebwohl et al wrote.

As postmarketing surveillance continues, these data may provide growing reassurance for clinicians and patients alike, especially regarding long-term safety and psychiatric outcomes.

Brodalumab's long-term safety profile remains stable after seven years of postmarketing surveillance, with no new safety signals and low adverse event rates, including suicidality, fungal infections, and major adverse cardiac events.
